Namajunas was born on Saints Peter and Paul, 1992, in Milwaukee, Wisconsin, to Lithuanian parents. Rose was named after her great-grandmother Ro Gotalkait Namajnien, the husband of an independent Lithuania officer. He was assassinated by the Soviets near his home in Kaunas, Lithuania's temporary capital. Rose Namajunas visits Lithuania on a regular basis and understands the Lithuanian language because she communicates with her grandparents in it.
Her father, she claims, suffered from schizophrenia, which caused complications and hardships during her childhood. Namajunas began learning Taekwondo at the age of five. She received her poom belt at the age of nine. She then went on to learn karate and jiu-jitsu. She began training in kickboxing and mixed martial arts with Duke Roufus at Roufusport while in high school, and she or he was also a senior-year wrestler at Milwaukee High School of the Humanities.

Namajunas began his MMA career as an amateur in 2010. At Minnesota Martial Arts Academy, she studied under Greg Nelson. She had a perfect amateur record of 4-0, with two wins by TKO and two by decision. Both finishes occurred in the first round.

On December 11, 2013, it was announced that Namajunas, along with ten other strawweight fighters, had been signed by the Ultimate Fighting Championship (UFC) to compete on season 20 of the Ultimate Fighter, which would crown the first-ever UFC strawweight champion. Gilbert Melendez selected Namajunas as the fourth pick. In the preliminary round of the tournament, she submitted Alex Chambers with a rear-naked choke. Namajunas faced Joanne Calderwood in the quarter-finals. She won the match by kimura in the second round.
Namajunas fought Angela Hill at UFC 192 on October 3, 2015. Namajunas knocked Hill out with a punch after some back-and-forth striking exchanges on the feet. Namajunas quickly secured a rear-naked choke as Hill stood up, submitting Hill in the first round.
